Preclinical Study of Cancer Metastasis Inhibition by Therapeutic Agents

Mouse Survival Study in a Systemic Metastasis Model. Cancer cells are injected into the cardiac left ventricle to create a model of systemic metastasis occurrence with involvement of major organs (liver, lung, brain, adrenals, bones, kidney, testes, etc) and the Kaplan Meier curves are determined in vehicle and drug-treated mouse groups.

Organ-Specific Metastasis Study. We compare antimetastatic action among different target organs and determine organ-specificity level.

Metastasis Implantation and Development Study: We comparatively evaluate the relative action of therapeutic agents on metastasis implantation (metastasis preventive agents) and development (metastasis growth inhibitors) stages.

Drug Synergism Study: Cellular signaling networks contain redundant pathways and multiple contributing cell types so that modulating a single pathway or cell type in the system cannot achieve the desired therapeutic effect. We assay for combination of the tested drug with another acting at a different level to target multiple pathways simultaneously.

Metastasis Dormancy versus Recurrence Study: Frequently, dormant or slow-growing subclinical metastases occur in cancer patients. We assay for drug effect on occult experimental micrometastases at prevascular and angiogenic stages, measuring 50-200 µm and 200-800 µm in diameter, respectively.

Microenvironment-Promoted Metastasis Study. We test for drug effect on metastases induced by selected pathophysiological processes that are concomitant to cancer progression in certain patient groups. The following experimental models are developed prior to cancer cell injection:
